• A3-Chol@NP induces M1 macrophage polarization and boosts tumor cell phagocytosis. • A3-Chol@NP colocalized with mitochondria in macrophages to generate mtROS. • The mtROS activates NF-κB-iNOS and IRF5-IL-23 pathways, driving M1 polarization. • A3-Chol@NP efficiently suppressed tumor growth via macrophage-mediated immunotherapy.
