• Carrier-free MTBN enables targeted eradication of intracellular S. aureus. • MTBN achieved 99.99% and 99.9% intracellular bacterial killing in vitro and in vivo, respectively. • MTBN enables cascade delivery, lysosomal escape, and bacterial penetration. • Synchrotron nano-tomography directly verified MTBN entry into bacteria in 3D.
