Neurofibromatosis type 1 (NF1) is an inherited disorder that can cause multiple nerve sheath tumors and neuropathic pain. Raut et al. found a potentially targetable source of neuropathic pain in NF1 that was independent of and before tumor development. In a mouse model of NF1, nerve-ensheathing Schwann cells produced and released more of the neurotrophic factor GDNF, which activated its receptor on peripheral sensory neurons, heightening their sensitivity to touch. This sensitivity was reduced by blocking GDNF production with an inhibitor that is already FDA approved to treat NF1 tumors, suggesting that it may be repurposed to treat neuropathic pain in patients. —Leslie K.
