Marine fungal-derived natural products against prostate cancer: A comprehensive review of discoveries and mechanisms

Prostate cancer, the most common malignancy in men, continues to show rising trends in both incidence and mortality. Despite significant advances in treatment and management, the rapid progression of prostate cancer and the treatment of advanced-stage disease remain formidable challenges. In recent years, marine fungi have gained attention for their ability to produce structurally novel natural products with potent antitumor activities, making them promising sources for new anticancer agents. This review provides a comprehensive summary of 153 natural products with anti-prostate cancer activity that have been isolated from marine fungi up to September 2025, with a focus on their sources, structural diversity, synthetic approaches, and mechanisms of action. These fungi were obtained from diverse marine substrates such as sponges, algae, sediments, and mangrove plants. The compounds exhibit significant structural variety, including polyketides, nitrogenous compounds, terpenoids and steroids. This review provides valuable insights into the discovery of marine-derived drugs for prostate cancer and elucidates potential therapeutic mechanisms.
