• MCART1 deficiency exacerbates mitochondrial dysfunction, manifested by loss of mitochondrial membrane potential, ATP depletion, and ROS accumulation. • MCART1-mediated mitochondrial NAD+ import is essential for sustaining mitochondrial function and cell survival under Parkinsonian stress. • Loss-of-function mutations in the key NAD+-binding sites abolish the protective activity of MCART1.
