• A novel 3D model has been developed to simulate the alveolar–vascular barrier in vitro. • PM2.5 exposure increased EGF and TNF-α expression in pulmonary epithelial cells. • EGF and TNF-α secreted by pulmonary epithelial cells transmit signals to vascular endothelial cells. • EGF and TNF-α regulate Nrf2 and CYP1A1expression in endothelial cells via the PI3K/Akt pathway. • Reveal the molecular link between alveolar injury and vascular endothelial dysfunction.
