• NAc MeCP2 knockdown increases operant cocaine self-administration at the unit dose tested. • shMeCP2 mice show higher cocaine infusions and active lever responding than controls during the maintenance phase and under a progressive-ratio schedule. • NAc MeCP2 knockdown does not alter baseline locomotion, anxiety-like behavior, or food-reinforced operant performance. • CPP-assessed cocaine-conditioned reward does not differ between groups under matched passive cocaine exposure (single dose).
