• FoxE3-Cre deletion of Meis1/2 from E10.5 reveals post-placodal lens roles. • E12.5 marks onset of epithelial disorganization with lens hypoplasia in double KOs. • Elevated apoptosis and aberrant fiber-cell cycling impair lens growth. • Meis1/2 maintain Pax6/FoxE3/Sox1; Sox2 becomes ectopic in lens epithelium. • Central ZO-1 loss and αSMA induction precede triangular, detached lenses. • Single KOs diverge: Meis1 variable “big eye”; Meis2 largely normal.
