• Dihydroartemisinic acid-produced Saccharomyces cerevisiae strain was constructed. • AaDbr2-ALDH1 fusion strategy increases DHAA/AA ratio from 0.6 to 4.2. • Cofactor and P450 complex optimization improved DHAA production by 9.3%. • Carbon source regulation decouples growth from DHAA production. • Fed-batch fermentation achieved the highest DHAA titer to 6.8 g/L to date.
