• LC-MS/MS method was developed for WKYMVm quantification in biomatrices. • LC-QTOF-MS with MSE identified sequentially truncated metabolites of WKYMVm. • Kinetic modeling pinpointed the Lys-Tyr bond as the primary metabolic soft spot. • Integrated modeling projected the limited oral dosing feasibility for WKYMVm.
