Metabolic regulatory nodes of the inflammasome and inflammatory cell death

Metabolic pathways actively instruct inflammasome signaling and the execution of inflammatory cell death programs, rather than serving solely bioenergetic roles. Distinct metabolic circuits converge on shared regulatory nodes; however, the mechanisms by which individual metabolites selectively control specific cell death modalities remain incompletely defined. Recent studies reveal extensive crosstalk among pyroptosis, necroptosis, PANoptosis, and ferroptosis, coordinated by the cellular metabolic state. The concept of ‘metabolic checkpoints’ integrates nutrient sensing and redox balance with inflammasome signaling and cell death execution. Defining context-specific metabolic dependencies will be critical for targeting immunometabolism in inflammatory diseases and cancer.
