• Micro-FTIR tracks AMSC dynamic osteodifferentiation despite high variability. • A protein/lipid spectral signature describes high proliferation conditions. • Inter- and intra-sample heterogeneity describes osteodifferentiating cells. • An osteo-specific spectral signature is marker of osteodifferentiation extension. • Differentiation increases helical proteins, lipids & 1030 cm−1 phosphate band.
