Midline assembloids reveal regulators of human axon guidance

The mechanisms of cell specification and axon guidance of the human nervous system during development remain to be fully elucidated, in part due to the paucity of model systems. Onesto et al. developed human floor plate organoids from stem cells and integrated them with spinal cord organoids to create human midline assembloids. Using this system, the authors profiled the secretome of human floor plate cells and discovered proteins that were not found to be expressed by the mouse floor plate. Functional work revealed new players in axon guidance during midline crossing. This approach is well suited to revealing distinctive aspects of human commissural axon guidance that might be otherwise difficult to approach experimentally. —Mattia Maroso
