• ZIF-8 exhibits formulation-dependent toxicity to earthworms, with methanol-dispersed particles showing the lowest LC50 value. • ZIF-8 induces severe reproductive damage, including clitellar atrophy and germinal epithelial degeneration. • Mitochondrial ultrastructural disruption is associated with ZIF-8-induced reproductive toxicity. • Formulation characteristics and mitochondrial toxicity are critical for nanomaterial environmental risk assessment.
