• Unsymmetrical pyridinophane–picolinate ligands and their Mn(II) complexes are reported. • Methylation of secondary amine enhances chelator kinetic inertness. • Additional chelating arms improve stability but reduce kinetic inertness. • Relaxivity differences are not solely due to molecular weight. • One Mn(II) contrast agent shows optimal renal excretion in vivo.
