• Modified β-glucans increase saponin and FCS accumulation in A. japonicus. • Modified β-glucans up-regulate saponin and FCS biosynthesis genes. • Modified β-glucans up-regulate TLR-MAPK axis genes and MAPK (p38/ERK) proteins. • MAPK blockade reduces β-glucan-induced saponin and FCS accumulation. • TLR-MAPK axis associates immune responses with saponin and FCS accumulation.
