Abstract

Abstract

In oncology, given the rising mortality rate, novel strategies aimed at developing effective and less toxic cancer treatments that are more selective. Thermal therapies such as magnetic hyperthermia therapies (MHT), photo-thermal therapies (PTT), and microwave therapies (MW) based on inorganic nanoparticles (iNPs) are emerging treatments which can induce cell death by apoptotic or necrotic processes, as well as by reshaping tumor microenvironment (TME), making cancer cells more sensitive to the drug effect. Metal-organic frameworks (MOFs) have also attracted extensive interest as drug carriers due to their large porous structures, huge surface area, and adjustable composition. This review will focus on MOF and iNPs combination, to achieve externally triggered drug release and hyperthermia based on one of the three modalities. From synthesis to applications, here we review the main materials developments in MOF and iNPs-MOF composites specifically designed for hyperthermia applications, their potential for improving pharmacokinetics and diagnostic aids, with an outlook to advance multimodal therapies and biocompatibility.
