• Five core NLRP3 inflammasome genes from Trachinotus ovatus were cloned and characterized. • Expression profiles of the genes were examined in liver, spleen, head kidney and intestine post S. agalactiae challenge. • ToNLRP3 was distributed in the cytoplasm, and ToASC was co-localized with ToCaspase-1 with obvious interaction. • Nigericin and ATP (NLRP3 inflammasome inducer/agonist) markedly upregulated ToNLRP3, ToASC and ToCaspase-1 expression.
