• First NIR fluorescent nanoprobe (D-CRP) for real-time Cu²⁺ monitoring in diabetic wounds, addressing a critical gap in understanding copper dysregulation in diabetes. • DSPE-PEG2000 encapsulation enhances biocompatibility and stability while preserving Cu²⁺-responsive NIR fluorescence (740 nm emission). • In vivo imaging reveals elevated Cu²⁺ levels in diabetic wounds and urine, correlating with impaired healing—a previously unexplored biomarker.
