• Breaking through the limitations of traditional neuroprotective studies, the activity of 4-HBd is directly related to the pathological process of HT (rather than simple cerebral ischemia), providing a new candidate molecule for the prevention and treatment of HT. • It is revealed that the active ingredient 4-HBd of Gastrodia elata reduces the post-HT in IS by regulating the PI3K-AKT pathway, which expands the modern pharmacological connotation of the ' Tongluo ' effect of Gastrodia elata. • By integrating omics data, a ' gene-metabolite-pathway ' regulatory network was systematically constructed, rather than a single omics or isolated pathway analysis, from molecular docking virtual screening to multi-level verification of animal/cell experiments to elucidate its multi-dimensional anti-HT mechanism. • It breaks through the single cognition of the mechanism of Gastrodia elata components in traditional research, and provides a novel candidate molecule and mechanism framework for the natural drug prevention and treatment of HT.
