• Spherical nucleic acids (SNAs)-driven catalytic hairpin assembly (CHA) enables multiplexed imaging of miR-155 and miR-210 in living RAW264.7 cells. • Enzyme-free signal amplification based SNAs-CHA with LODs of 0.033 nmol/L (miR-155) and 0.026 nmol/L (miR-210) in vitro, respectively. • Low-dose exposure (0.05–2 μmol/L) to DON, 3-Ac-DON, and 15-Ac-DON induces time-dependent upregulation of miR-155 and miR-210 in vivo. • Dual-miRNA imaging provides a novel strategy for early screening of immunotoxicity induced by DON and its acetylated derivatives.
