N-Halosuccinimides (NBS or NCS) or trichloroisocyanuric acid (TCCA) were employed as catalysts for the hydrolytic cleavage of glycosidic bonds in digitalis derivatives, including digoxin and digitoxin. A simple, scalable, and operationally straightforward protocol was developed for the preparation of genins from digitalis steroids using commercially available, non-corrosive catalysts in a hydroalcoholic medium. This method affords clean products in excellent yields (96-99%), with purity confirmed by NMR and HRMS after simple filtration and solvent evaporation, thereby eliminating the need for chromatographic purification.
