Nanodiamonds (NDs) have garnered significant attention due to their unique potential, including displaying excellent bacterial resistance and favourable interactions with mammalian cells. Here, we show multifaceted biocompatibility of NDs both in vitro and in vivo. The NDs show enhanced mammalian biocompatibility via the key markers of proliferation and differentiation of mouse bone stromal (Kusa4b10), human kidney (HEK293), and mouse dermal fibroblast (L929) cells. Importantly, high surface area, hydrophilicity, and chemical stability contribute to their antibacterial efficacy against various bacterial strains. Our findings reveal that Escherichia coli is more sensitive to NDs than Staphylococcus aureus although both E. coli and S. aureus show a significantly reduced viability of 8% with exposure to 10 mg/mL of ND. To further confirm their potential in vivo, zebrafish embryos exposed to NDs were observed to display normal development and healthy spine formation, indicating the absence of teratogenic effects. These results position NDs as promising candidates for biomedical applications, including drug delivery, biosensing, and tissue engineering.
