• Tetramine poisoning frequently leads to epilepsy, which has complex mechanisms and poor treatment outcomes. • This study identified HIF1A, HSP90AB1, MMP9, SRC, and STAT3 as core targets regulating tetramine-induced epilepsy. • The HIF-1, PI3K-Akt, Ras, Toll-like receptor, chemokine, and neurotrophin signaling pathways appear to be critical in the regulation of tetramine-induced epilepsy. • Network toxicology methods have indicated that tetramine may induce epilepsy through multiple targets and signaling pathways.
