• Sleep quality differs in youth, adolescents, and young adults with a high risk for depression. • REM sleep neurophysiology and sleep spindle density differ by depression risk. • Poor sleep was associated with differences in white matter integrity, which in turn predicted future depressive symptoms. • Reward related brain activity in the dorsomedial prefrontal cortex and resting-state salience network activity mediated the relationship between sleep and future depressive symptoms. • Substantial heterogeneity, but low-to-moderate risk of bias, was observed.
