• CCI downregulates neuropeptide S (NPS) and its receptor expression in the LH. • Intra-LH infusion of NPS produces antinociceptive effect in CCI rats. • NPS receptor antagonist SHA-68 (intra-LH) exacerbates neuropathic pain. • NPS activates LH-vlPAG orexinergic pathway to produce antinociception. • NPS treatment restored injury-induced neurochemical imbalance in the vlPAG.
