• Elevated circulating endocan levels contribute to blood pressure elevation and vascular dysfunction in hypertension. • Endocan induces endothelial dysfunction by TNF-α–dependent inhibition of eNOS phosphorylation and NO bioavailability. • Endocan neutralization attenuates hypertension and improves vascular function in Ang II–induced hypertension mice. • Endocan may serve as a promising therapeutic target for managing hypertension and associated cardiovascular complications.
