• Nickel exposure in a closed-cycle NFT system caused a rapid, dose-dependent reduction in lettuce growth and yield. • Even moderate Ni concentrations (75 µM) significantly decreased water and nutrient use efficiency. • Nickel accumulation was predominantly localized in roots, confirming root sequestration as a primary detoxification mechanism. • Prolonged exposure increased Ni translocation to leaves, leading to critical accumulation in edible tissues. • Antioxidant enzymes (CAT, POD, SOD) and nitrogen metabolism (NR, NiR) showed cultivardependent responses to Ni stress.
