• OsCYP51H9, an ER-localized P450 in rice, is predominantly expressed in reproductive and vascular tissues. • Knockout of OsCYP51H9 disrupts phytosterol/BR metabolism, causing dwarfism, poor roots, small grains, and erect leaves. • Overexpression of OsCYP51H9 under low nitrogen enhances root growth, nitrogen accumulation, grain yield, and upregulates nitrogen-responsive transcription factors OsWRKY69 and OsDREB1C/B. • OsCYP51H9 bridges BR-dependent development with nitrogen adaptation, presenting a promising target for high-yield, low-fertilizer rice breeding.
