• StcE, a bacterial mucinase, shows promise for transiently disrupting mucus barriers to enhance drug diffusion. • Mucolytic activity of StcE varies across porcine mucus samples, despite preserved enzyme integrity. • Zinc supplementation improves StcE function in degradation resistant mucus. • Cofactor availability, not degradation or inhibition, appears to be a determinant of StcE efficacy in mucus.
