• Oxygen tension reshapes how trophoblasts interpret TGF-β1 signaling. • Combined O₂-TGF-β1 control determines fibronectin abundance and assembly. • Fibronectin-integrin engagement regulates invasion and endothelial-like behavior. • Hypoxia disrupts this signaling–matrix axis in severe preeclampsia tissue. • Findings reveal fibronectin as an instructive determinant of trophoblast fate.
