• A novel palladium-catalyzed carbonylative cyclization is developed for the synthesis of quinazolinones using near-stoichiometric CO2. • A protocol delivers products in good yields with broad functional group tolerance, enabling one-step access to 13C-labeled quinazolinones • Mechanistic investigations combining control experiments and in situ 13C NMR verify the DBU-CO2 adduct and N-benzoylated intermediate as key active species.
