• Pantheon-DNA integrates adaptive NGS preprocessing with a robust encoding-decoding codec. • Modular pipeline: Chiron (demultiplexing), Hermes (addressing), Hephaestus (clustering). • Achieves ≥99.996% retrievability at 10× coverage under LER and HER in our tests. • Delivers faster decoding while reserving MSA for cases that require it, reducing computational time. • Scales to 90 MB in silico and is validated by a 1.59 MB synthesis-–sequencing experiment.
