• Three pharmaceutically acceptable salts of niflumic acid (NIF) with tromethamine, piperazine and triethanolamine were prepared and fully characterized. • Single-crystal structures of NIF-PIP (2:1, hexagonal R-3, hydrophobic channel) and NIF-TEA (1:1, monoclinic P21/n, hydrophilic 3D wavy network) are reported. • All three salts significantly improved equilibrium solubility and intrinsic dissolution rate in water and pH 6.86 buffer. • NIF-TEA exhibited the highest enhancement: ∼1145-fold solubility and ∼3212-fold IDR in water, correlating with its open hydrophilic network.
