• Physiological oxygen tensions profoundly alter anticancer drug responses. • Dihydroartemisinin showed the strongest activity across cancer cell lines. • Hypoxia enhanced apoptosis and oxidative stress induced by DHA. • Lipid peroxidation increased despite largely unchanged GPX4 expression.
