• PLA2G16 knockout sensitizes CRC to OXA in vitro and in vivo. • PLA2G16 deficiency is associated with altered autophagy-related markers in response to OXA. • Transcriptomic and lipidomic analyses suggest an association of PLA2G16 with lipid metabolism and autophagy-related pathways. • PLA2G16 may be a potential target for improving oxaliplatin response in CRC.
