• RPmFREP1 selectively inhibits bacterial growth, with strong activity against S. aureus and V. parahaemolyticus. • RPmFREP1 enhances the agglutination of multiple Gram-negative and Gram-positive bacterial species in the presence of Ca2+. • RPmFREP1 contributes to antibacterial defense, and antibody blockade markedly reduces Pinctada fucata martensii survival after V. parahaemolyticus challenge.
