• A dual-miRNA electrochemical sensor is developed based on a polygonal nucleic acid frame-driven DNA nanomachine. • The dual detection of miRNA-21 and miRNA-105 is achieved with detection limits at the aM level. • Substrate-free AuPd@CoMOF electrocatalysis eliminates exogenous analytical errors. • Spatial confinement shortens reaction time by 55% and enhances current response by 63%. • The sensor discriminates metastatic from non-metastatic breast cancer cells and matches qRT-PCR in clinical serum.
