• PS-NPs exposure impairs placental nicotinamide metabolism and mitochondrial energetics. • PS-NPs triggers NMNAT3 downregulation, NAD+ depletion, and ferritinophagy-driven ferroptosis. • NMNAT3 overexpression rescued PS-NPs-induced defects by inhibiting ferritinophagy-mediated ferroptosis. • NAM supplementation restores iron homeostasis, suppresses ferroptosis, and rescues adverse pregnancy outcomes.
