Precision insect control using programmable modular phage therapy platforms

Abstract

Spodoptera frugiperda, a major global pest, is resistant to several chemical insecticides via gut symbiont detoxification metabolism. This reduces the effectiveness of traditional control and increases insecticide residues and ecological risks, thereby threatening sustainable agriculture. Phage SP-1 (a newly isolated phage strain deposited at the China Center for Type Culture Collection (CCTCC) under accession number CCTCC M 20241620) targets and lyses the symbiont Enterococcus casseliflavus that mediates detoxification metabolism of insecticides, thereby restoring sensitivity to chlorantraniliprole. Zein-based nano-insecticide co-encapsulates SP-1 and chlorantraniliprole, withstands 60°C heat and UV. Compared with free chlorantraniliprole, the nano-insecticide improves Spodoptera frugiperda control by 17%, reduces soil residues by 82.4%, and is safe for the environmental microbial community and maize. The Modular Phage-based Symbiont Targeting platform enables flexible component swapping, offering a sustainable solution for resistant pests. The Modular Phage-based Symbiont Targeting system targets gut symbionts of Spodoptera frugiperda, rapidly reversing insecticide resistance. It reduces chemical pesticide use and residue while increasing control efficacy, offering a precise, adaptable, and eco-friendly solution for sustainable management of resistant agricultural pests.
