• A novel, low-cost expression system was established using Daphnia magna to produce and accumulate recombinant proteins in eggs. • Vitellogenin signal peptide (VtgSP) and beta-sheet domain (BS) were identified and used to mediate secretion into hemolymph and uptake into eggs. • The system enables simple purification by accumulating proteins in easily isolated eggs and represents the first in vivo analysis of vitellogenin-based transport in Daphnia.
