• A specific scarlet ortholog is highly expressed in the locust brain and antennae. • scarlet RNAi knockdown disrupts olfactory attraction to plant volatiles. • scarlet knockdown results in a metabolic shift for 3-OHK and XA metabolites. • XA recapitulates the olfactory attraction impairment in migratory locusts. • XA suppresses neurophysiological activities in the antennae and brains of locusts. • R-loop Scarlet-RL at the scarlet locus suppresses scarlet transcription.
