• RPA-flocculation enabled rapid detection of Paramarasmius palmivorus in oil palm. • Species-specific primers amplified target DNA within 15 min at 42 °C using RPA. • No cross-reactivity was observed with 36 non-target fungal species or host DNA. • The assay achieved a detection limit of 10 pg/μL genomic DNA. • Simple DNA extraction to RPA-flocculation workflow completed within 70 min.
