Rapid selection of transgenic mammalian cells via diphtheria toxin resistance

Abstract

We describe an efficient method to select transgene-positive mammalian cells. We engineered a fusion protein to efficiently protect cells from diphtheria toxin (selecDT). The novel selection marker selecDT is efficiently integrated along with large transgenes. Simple nonviral transfections were combined with overnight selection of human cells. A broad selection window for many common cell lines minimizes optimization. Efficacy was shown in the most common producer cells HEK293 and CHO. Fast and efficient selections minimize consumables needed for cell line creation. The system is orthogonal to existing antibiotics and may be further expanded with other immunotoxins.
