• A series of amino-functionalized xanthene dyes (AXN 1–3) was developed through rational electron donor engineering. • The optimized dye AXN-3 dye exhibits large Stokes shifts (136 nm) and efficient near-infrared (NIR) emission (675 nm). • A NE-activated NIR fluorescent probe (AXN-NE) was developed, enabling in vivo mapping of neutrophils recruitment during inflammatory process.
