Abstract

This review focuses on three core strategies in the field of quinoline synthesis, classified by the number of bonds formed into single-bond cyclization, two-bond formation, and multicomponent three or more bond formation. The latest advances and key advantages of each method are systematically summarized. The results indicate that single-bond cyclization relies on intramolecular pre-functionalized precursors to achieve precise bond formation, making it suitable for highly selective targeted synthesis. Compared to single-bond cyclization, tandem cyclization reactions significantly shorten synthetic routes and demonstrate good potential for scale-up applications. Multicomponent reactions, characterized by the construction of multiple bonds in a single step, enable efficient utilization of starting materials and rapid introduction of structural diversity, aligning with the demands of green chemistry and sustainable synthesis. These three complementary strategies collectively drive the evolution of quinoline synthesis toward high efficiency, low consumption, and high selectivity, providing methodological support for the widespread applications of quinoline derivatives in pharmaceuticals, agrochemicals, and functional materials.
