• Redox-responsive lipid nanoparticles enable spatiotemporally controlled cargo release by coupling disease-specific GSH/ROS gradients with programmable structural transformations. • Rational integration of responsive motifs into LNP improves endosomal escape, delivery efficiency, and therapeutic potency of gene or drugs. • Emerging designs and targeting strategies position redox-LNPs as versatile platforms for developing precision medicine across cancer and non-cancer indications.
