Rhizosphere inoculation with Streptomyces pactum is associated with sugar-related fruit quality and rhizosphere variation in goji berry (Lycium barbarum L.)

Goji berry production is increasingly constrained by root rot and continuous cropping related soil problems, yet the relationships among rhizosphere conditions, microbial community variation, and fruit quality remain unclear. This study evaluated whether rhizosphere inoculation with Streptomyces pactum Act12 (Act12) was associated with changes in rhizosphere properties, microbial communities, and fruit quality-related traits under field conditions. A field experiment compared Act12 inoculation with a non-inoculated control. Rhizosphere soil physicochemical properties, plant growth, yield, and fruit nutritional traits were measured. Full-length 16S and 18S rRNA amplicon sequencing was used to characterize bacterial and eukaryotic community composition. Community environment relationships were assessed using ordination, variance partitioning, Mantel tests, and correlation based network analysis. Act12 inoculation was associated with higher rhizosphere soil organic matter and lower residual levels of available nitrogen and potassium. Inoculated plants also showed greater vigor, improved canopy-related traits, larger fruits, and higher yield. Fruit nutritional responses were mainly reflected in carbohydrate, with increased soluble sugars and a moderate increase in polysaccharides. Act12 inoculation was also associated with higher microbial richness and diversity, and with shifts in bacterial and eukaryotic community composition. Integrative analyses further showed that soil organic matter and nutrient indices were significantly associated with dominant microbial taxa and sugar-related fruit traits. Under field conditions, Act12 inoculation was associated with changes in rhizosphere soil properties, microbial community composition, and carbohydrate related fruit traits in goji berry, supporting its potential as a rhizosphere management input in orchards.
