• rSiglec-10(V set) enhanced the oncolysis through CD24-Src-ERK pathway. • OVs-rSiglec-10(V set) therapy showed superior outcomes in tumors with high CD24. • OVs-rSiglec-10(V set) therapy reshaped the immunosuppressive tumor microenvironment. • OVs-rSiglec-10(V set) plus anti-PD-L1 showed a synergistic effect.
