Abstract

• Deletion of Runx3 in thymic epithelium leads to loss of Aire + mTECs and an increase in immature Ccl21a-expressing mTECs. • Runx3-deletion leads to loss of tissue specific antigen expression, autoimmunity, and increased escape of autoreactive T cells from the thymus. • Runx3 affects signaling pathways important for inducing Aire + mTEC development, including CD40 and NF-kb.
